This protocol describes the methodology for determining the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) of antimicrobial compounds against halophilic archaea using a 96-well microtitre plate assay. MIC values are visually determined based on growth inhibition, while MBC values are confirmed through subculture on Payne’s medium (PM) agar. The method follows NCCLS guidelines (NCCLS, 2000), with modifications tailored for haloarchaeal growth conditions.
